Depotformen
zur parenteralen und peroralen Applikation
von Peptiden und Proteinen

Arzneistoffe mit Peptid- bzw. Proteinstuktur sind hochpotente Pharmaka. Ihre im Vergleich zu herkömmlichen Arzneistoffen völlig andersartigen physikochemischen Eigenschaften sowie die damit verbundenen Stabilitäts- und Bioverfügbarkeitsprobleme erfordern neue galenische Konzepte zur Herstellung geeigneter Arzneiformen.Im Rahmen des Forschungsprojektes werden:

  • Hilfsstoffe und Verfahren zur Herstellung parenteral bzw. per os applizierbarer Depotformen gezielt variiert und ihr Einfluss auf Beladung, Freigabekinetik, Freigabemechanismus, Struktur und Aktivität bzw. Antigenität verschiedener Modellpeptide und -proteine untersucht
  • neue Verfahren zur Herstellung mikro- und nanopartikulärer Arzneiformen entwickelt
  • Therapieeffekte untersucht

Eines der NIR-Geräte am Institut

 

Steigende gesetzliche Anforderungen an die Kontrolle von Identität und Qualität pharmazeutischer Produkte erfordern Analysentechniken, die einfache, sichere, kostengünstige und zerstörungsfreie Untersuchungen ermöglichen. Um eine lückenlose Prüfung ganzer Chargen zu ermöglichen, sollte eine online oder inline Anwendung möglich sein.

Die Spektroskopie im Bereich des Nahen Infrarot erfüllt diese Anforderungen weitestgehend. Sie ermöglicht automatisierte, schnelle, preiswerte, zerstörungsfreie und damit nicht nur stichprobenweise sondern vollständige Untersuchungen ganzer Chargen. Aus NIR-Spektren lassen sich außerdem gleichzeitig Aussagen über unterschiedlichste Parameter einer Arzneiform ableiten (siehe unten).

Trotz der Aufnahme in das Europäische Arzneibuch 1997 konnte sich die NIR-Spektroskopie, vermutlich nicht zuletzt aufgrund der komplizierten chemometrischen Auswerteverfahren und des beträchtlichen Aufwandes bei der Ersterstellung der nötigen Kalibrationen, bislang noch nicht in großem Maßstab durchsetzen. Im Arbeitskreis sollen Grundlagen erarbeitet und pharmazeutische Anwendungsmöglichkeiten erforscht werden.

Bisherige Untersuchungen an Kapseln und (überzogenen) Tabletten waren vielversprechend. So konnten aus NIR-Spektren Aussagen über folgende Parameter gewonnen werden:

  • Filmdicke, Morphologie und Mikrostruktur von Filmüberzügen auf Tabletten
  • Porosität, Härte, Zerfall und Wirkstofffreisetzung von Tabletten und Filmtabletten
  • Mikrostruktur, mechanische Festigkeit, Löslichkeit und Lösegeschwindigkeit von Gelatinekapselhüllen

In weiteren Projekten sollen bioabbaubare monolithische oder partikuläre Arzneiformen auf Basis von Polylaktiden bzw. -glykoliden mittels NIR-spektroskopischer Methoden untersucht werden. Ziel ist eine Charakterisierung ihrer Zuammensetzung und der biopharmazeutisch relevanten Eigenschaften. Im weiteren Verlauf ist an in vitro- und in vivo-Untersuchungen der Freisetzungseigenschaften peptidhaltiger Depotarzneiformen gedacht.
